This study was conducted to determine if it was possible to vary the proportion of bull sperm with Y chromosome by using bovine serum albumin “BSA“ in the following three methods: Double centrifugation in BSA (M1); centrifugation and migration in BSA (M2); Double migration in BSA (M3). The samples obtained, were stored for 20-24 hours at 5°C; then frozen in liquid nitrogen. The variation in the proportion of Y chromosome (f-body) spermatozoa was determined using staining with quinacrine dihydrochloride. No significant difference was observed (P> 0.05) in the percentage of (f-body); 48.41 (± 5.93)% for the control; 46.15 (± 5.86)% for (M1); 45.82 (± 5.47)% for (M2) and 47.18 (± 4.92)% for (M3). The spermatozoa retrieval rate differed among the methods evaluated (P <0.01), with 50.69% for (M1), 36.23% for (M2) and 21.41% for (M3). The effect of ASB on sperm characteristics was significant (P <0.01). The sperm vitality was reduced from 64.24% in the control to 42.53% for (M1), 37.23% for (M2) and 27.23% for (M3). Motility was reduced from 83.11% in the control to 48.11%, 31.89% and 23.11% for (M1), (M2) and (M3) respectively. The percentage of sperm with functional membrane was reduced from 64.62% in the control to 42.74%, 42.58% and 39.63% in (M1), (M2) and (M3) respectively. The percentage of live sperm acrosome unreacted was 58.12% in the control; 35.10% in (M1); 30.16% in (M2) and 20.48% in (M3). It is observed that freezing bull sperm in liquid nitrogen reduced the characteristics evaluated making them unviable.
