The present work consisted in the evaluation of the maturation of oocytes in vitro through the technique of fixation during metaphase II from post-mortem creole cows. Oocytes were recovered through the follicular aspiration technique of ovaries collected in slaughterhouse, and classified by categories and then matured in TCM 199 medium for 24 hours at 38ºC in an atmosphere of 5 percent CO2 and 90 percent humidity. At the end of the maturation period, the oocytes were fixed in ethanol: acetic acid (3/1) for 48 hours and in cooling 4 ° C. Then these were stained with aceto orcein to evaluate the nuclear stage under inverted lens microscope. The percentage of oocytes that reached the stage of metaphase II was 57.78 percent, while those not determined were 17.52 percent and in prophase and metaphase I, were 7.62 and 17.08 respectively. The test for the statistical evaluation was chi square, with 19 plates evaluated. Of the undetermined oocytes (ND), plate 11 was significant, with an undetermined oocyte (ND) of 6 observed (p = 0.07). Of the oocytes in metaphase I (MI), plate 17 was significant, with 2 oocytes in metaphase I (MI) of 14 observed (p = 0.001). Of the oocytes in metaphase II (MII) according to the chi square test, plate 5 and 7 were statistically significant, with a number of oocytes of 15 of 25 in plate 5, 12 of 20 in plate 7 (p = 0.001).
