Cloning and protein expression in heterologous systems are very useful tools for the study of viral proteins. In this work, an in vivo cloning strategy was applied using the budding yeast Saccharomyces cerevisiae, as an efficient and low-cost method to clone the complementary DNAs (cDNAs) from the Tilapia Lake Virus (TiLV), which is associated to worldwide massive mortalities of tilapia Oreochromis niloticus including Peru. Complementary DNAs synthetized from infected fish tissues allowed the cloning of all the viral segments with high efficiencies; while the bioinformatic analysis allowed the elucidation of a close phylogenetic relationship with a previously reported Ecuadorian strain responsible of syncytial hepatitis outbreaks since 2012.
