The present research work was carried out in the National Semen Bank of the National Agrarian University La Molina-Lima, with the objective of evaluating the effect of the addition of cholesterol-loaded methyl-β-cyclodextrin (CLC) on the characteristics microscopies of thawed ram semen, using 4 rams (Asblack breed) between 1 and 2 years of age. Thirty ejaculates were collected by the artificial vagina method and immediately transferred to the laboratory to evaluate their macroscopic and microscopic characteristics. After a 1:1 predilution, the semen was divided into 03 groups and the CLC (CholesterolLoaded Cyclodextrin) were added (0mg of CLC / 120 million sperm = T1, 1 mg of CLC / 120 million sperm = T2 and 2 mg of CLC / 120 million sperm = T3). The samples were incubated for 15 minutes at laboratory temperature (22ºC), the dilution was concluded and finally the temperature decrease was made up to 5 ° C in 2 hours plus a time of 18 hours of stabilization. The freezing of the semen was done in 0.5 ml straws (30 x 10 6 spermatozoa) using nitrogen vapors for 6 minutes (-120°C) to be submerged in liquid nitrogen (-196°C) and stored in cryogenic tanks. The defrosting of the straws was performed at 38ºC for 15 seconds and the motility, vitality, abnormalities and functionality of the spermatic membrane were evaluated. The post-thawing results show improvements with the use of cholesterol in the cryopreservation of ram semen, obtaining a motility of 65.34 percent, 68.68 percent, 75.27 percent, a viability of 56.58 percent, 60.70 percent, 66.23 percent, abnormalities of 9.60 percent, 10.13 percent, 11.23 percent and membrane integrity of 50.54 percent, 56.4 percent and 63.71 percent for T1, T2 and T3 respectively; there being a statistical difference (p <0.05) between the treatments in all the variables, except in morphology. Therefore, it is concluded that the incorporation of cholesterol to the sperm membranes through the use of cholesterol-loaded cyclodextrins is an efficient way to increase the quality of semen sperm parameters in cryopreservation
