Neurodegenerative pathologies are part of a heterogeneous group of diseases that adversely affect the functioning of the central nervous system. They are characterized by gradual or progressive neuronal damage in specific brain regions, leading to cognitive decline, reduced functionality, and loss of personal independence. Among neurodegenerative diseases is Parkinson's disease (PD), which ranks as one of the most common movement-related neurodegenerative disorders worldwide, occupying the second position overall. In PD, there is a deficiency in the nigrostriatal pathway, specifically a deficit in the dopaminergic projection of this circuit, which is part of the pyramidal system responsible for motor regulation. This deficiency manifests as movement disorders. In PD, the degeneration of neurons in the substantia nigra results in the loss of dopaminergic inhibition and a relative excess of cholinergic activity, both of which are associated with the movement disorders characteristic of the disease. At the presynaptic terminal, dopamine must remain harmless to the neuron by being stored in synaptic vesicles. Additionally, neurons produce a protein called α-synuclein, and mutations in this protein disrupt its capacity for proper storage, further contributing to the pathological process.
