--- Extreme resistance to PVY virus is controlled by Ry genes and is effective against all strains in contrast to hypersensitive resistance, which is specific to a particular strain. In Solanum tuberosum ssp andigena, the responsible for this resistance is Ryadg gene located in the chromosome XI. This work aims to characterize at molecular level the region of chromosome XI where the gene Ryadg, that confers resistance to PVY, is located in order to identify molecular markers closely linked to this gen, the order and distance of these in this region and apply these markers in identifying new sources of resistance to PVY in the germplasm of Solanum tuberosum ssp andigena. Segregating progeny evaluated to determine the order and distance of the RySC3, M45 y M6 markers was obtained cross the Costanera variety, PVY resistant to possess the Ryadg gene in simplex dosage, and the susceptible variety LBr-43. The three markers evaluated in the progeny segregated in a 1:1 ratio, confirming the Ryadg allelic dosage of Costanera. Of 1089 evaluated were obtained four recombinant individuals for all three markers assessed and the most likely order of these markers is M6 flanked by RySC3 (to 0,18 cM) and M45 (to 0,18 cM). Evaluation of resistance to PVY in these recombinants indicated that the Ryadg could be located between M6 and M45 markers, or near of this region. By using the M45 and RySC3 markers in a population of 251 accessions of S. tuberosum ssp andigena, this work found that only the Pisha Milpo accession was positive for these markers, and also for the M6 marker. Subsequently, was confirmed their identity as S. tuberosum ssp andigena using a chloroplast marker and microsatellites. Finally, the greenhouse evaluation confirms the PVY resistance in Pisha Milpo accession. Keywords: PVY, S. tuberosum ssp andigena, Ryadg, resistance, molecular markers
