Genetic screenings that rely on protein overexpression are methods that use modern analysis to systematically explore gene functions in vivo. The present investigation involved the optimization of two screening protocols that used a genomic library as a source of genetic diversity. This library is formed by thousands of diferent fragments of genomic DNA from the yeast S. cerevisiae. In the first method, transformed colonies were transferred by replica plating, and the replicated plates were exposed to UV radiation or seeded in medium containing hydrogen peroxide. After the screenings, tests were carried out with UV radiation and hydrogen peroxide to confirm the positive phenotypes. A total of five plasmids were isolated in the screenings and their analysis identified genes involved in the cellular response to damaging agents. In conclusion, the optimized screening protocols allowed identification of genomic library plasmids that included genes involved in the response to cellular damage..
