The objective of this study was to determine the relationship between knowledge and practice of exclusive breastfeeding in primiparous mothers attending the Calzada Health Center, 2021-2022. This was a quantitative, basic study with a cross-sectional correlational design, using a sample population of 69 primiparous mothers attending the Calzada Health Center, who met the inclusion and exclusion criteria. The techniques used were a survey and observation, using a questionnaire (17 questions) and an observation guide (35 items) as instruments. As a result of demographic data, 30.4% of the primiparous mothers were between 26 and 34 years old; 59.4% were from rural areas, 79.7% were independent mothers, 37.7% had a high school education, 42.0% had a low level of knowledge and 91.3% showed inadequate breastfeeding practices. It was found that there is a negative relationship between the level of knowledge and practice of breastfeeding, according to the Pearson correlation calculated (n=63), which rejects the null hypothesis, concluding that there is a relationship between the level of knowledge and practice of breastfeeding in primiparous mothers, making it relevant to plan promotional activities to improve the knowledge and practice of breastfeeding among the population.
