ABSTRACT The main objective of this work was to apply the procedures of ovariectomy, sequenced vaginal cytology and quantitative determination of the estrous cycle in Mus musculus balb/c. Eighteen prepubertal female Mus musculus balb/c were randomly distributed into 3 experimental groups: negative control, positive control and sham. The specimens were ovariectomized, through a single dorsal surgical incision in the midline of the abdomen, followed by the identification of the ovaries and their removal. The sham group underwent a simulated surgical procedure without removal of the ovaries. After ovariectomy, all specimens received post-operative treatment with tramadol and chloramphenicol for 4 days, and rested for 14 days for full recovery. The negative control received physiological saline solution (PSS) at a dose of 10 mL/kg orally (PO), the positive control received estradiol valerate at a dose of 1 mg/kg PO, the sham group received SSF at a ratio of 10 mL/kg VO, all groups received the treatments for 15 days. Vaginal secretion samples were taken between 8 and 10 a.m. for 6 days in the pre- treatment and 7 days in the post-treatment, a smear of vaginal secretion and the respective staining (hematoxylin-eosin) were made with the samples. Then, the quantitative determination of the estrous cycles of all the groups was carried out, considering that the estrous cycle is made up of four phases; proestrus, estrus, metestrus, and diestrus, and that each phase is characterized by a predominance or absence of nucleated cells, cornified cells, and/or leukocytes. It was evidenced that the negative control group both in the pre-treatment and post-treatment showed a predominance of the right-handed phase; likewise, the sham group showed a cyclic estrous behavior, while the positive control group in the pre-treatment was similar to the negative control, with a predominance of right-handedness; and in the post - treatment, a predominance was observed in the proestrus phase.
