Title: Cuantificación específica de biomasa en biopelículas fúngicas mixtas de Trichoderma reesei y Aspergillus spp. mediante PCR

Abstract: Production of cellulases by mixed cultures of A. niger and T. reesei shows that enzymatic titers of total cellulases, endoglucanases and β-glucosidases can be higher in comparison with individual cultures. Although mixed cultures constitute a promising fermentation system, it has not been addressed how much of this enzymatic synergism depends on a certain proportion of fungal mixed biomass. In this work, biomass was specifically quantified in biofilms composed of A. niger and T. reesei, two filamentous fungi commonly used in the industrial field. Mixed biofilms were collected from 48 to 120 hours of growth and DNA was extracted for qPCR assays with specific primers for each fungus. These primers were designed from non-conserved regions of actin and β-tubulin genes of both A. niger and T. reesei. Specificity of these primers was tested both in silico and experimentally. Using these primers, a statistically significant correlation was obtained between qPCRcalculated biomass and dry weight biomass data. Mycelium proportion in biofilms did change over time: at 48 hours, 62% of A. niger and 38% of T. reesei; 72 hours, 53.5% and 46.5%; 96 hours, 84.5% and 15.5% and 120 hours, 95.9% and 4.1%, respectively, suggesting a competitive interaction between these two fungi, at the end of which A. niger replaced almost completely T. reesei mycelium. In conclusion, it is possible to perform a specific quantification of biomass by qPCR in mixed biofilms of filamentous fungi. In fact, the presented method could also be applied to any culture system, in order to study metabolic responses that fungi, or even other microorganisms, display when interact with each other

La producción de celulasas mediante cultivos mixtos de A. niger y T. reesei muestra que los títulos enzimáticos de las celulasas totales, endoglucanasas y β-glucosidasas pueden ser superiores en comparación con los cultivos individuales. Si bien los cultivos mixtos representan un sistema de fermentación prometedor, no se ha evaluado en qué medida la proporción de biomasa mixta influye en este sinergismo enzimático. En este trabajo se ha cuantificado de manera específica la biomasa en biopelículas mixtas compuestas por A. niger y T. reesei, dos hongos filamentosos comúnmente usados en el ámbito industrial. Las biopelículas mixtas fueron colectadas desde las 48 hasta las 120 horas de crecimiento. De ellas se extrajo ADN para realizar ensayos de PCR cuantitativa (qPCR) con cebadores específicos para cada hongo. Estos cebadores fueron diseñados a partir de las zonas no conservadas de los genes actina y β-tubulina de A. niger y T. reesei. Se comprobó la especificidad de los cebadores tanto in silico como experimentalmente. Utilizando dichos cebadores, se obtuvo una correlación estadísticamente significativa entre la biomasa determinada mediante qPCR y la registrada mediante peso seco. La proporción de micelio de cada hongo en las biopelículas mixtas mostró un cambio con respecto al tiempo: a las 48 horas, 62% de A. niger y 38% de T. reesei; 72 horas, 53.5% y 46.5%; 96 horas, 84.5% y 15.5% y 120 horas, 95.9% y 4.1%, respectivamente, lo que sugiere una interacción competitiva entre estos dos hongos, al final de la cual el micelio de A. niger desplazó casi por completo al de T. reesei. En conclusión, es posible realizar una cuantificación específica de biomasa mediante qPCR en biopelículas de hongos filamentosos. Además, el método desarrollado podría ser aplicado a cualquier sistema mixto de cultivo, con el fin de estudiar las respuestas metabólicas que los hongos, o incluso otros microrganismos, experimentan al interactuar entre sí
